Treating Patients with Neurological Diseases

Neupanex® is the result of 25 years of research by Lewis Clarke, M.D., Ph.D. As a rehabilitation physician with a Ph.D. in Neuropharmacology and Cell Biology, Dr. Clarke has studied the neurochemistry of neurologic function and dysfunction since 1976. When he began treating patients with debilitating neurologic diseases such as strokes, spinal cord injuries, Guillian-Barre, and degenerative neurologic processes such as MS, ALS, Alzheimer’s, and Parkinson’s, he began implementing interventional and neuroregenerative neuropharmacology. Here is a quote from one of his papers.

“The final frontier in medicine is the brain. It would appear that we will never comprehend the intricacies of its anatomical network, its chemistry, neuronal and glial cell biology, and the delicate balance of inhibition and facilitation and modulation, and its interaction and ultimate control with and of the rest of the body.  It is a symphony of literally millions of instruments. As incomplete as is our understanding of its function, even more so is our lack of comprehension of its pathologies (disease processes). Simplistic conceptualization of a stroke as a plumbing problem followed by ischemia (a clogged pipe followed by lack of blood and oxygen) with most of the blame and the hope hanging respectively on cholesterol and drugs that lower it, summarizes the current level of understanding both of the general public and of physicians as well. 

This should not be!  A casual search of the current literature reveals thousands of studies detailing neuropharmacology, mitochondrial oxidative phosphorylation and β-oxidation, DNA on/off switches, membrane and nuclear receptors, endoplasmic reticulum and mitochondrial interaction, excitotoxic neurotransmitters, free radicals, and inflammatory cascades. It is certain that these represent only a few of the many mechanisms involved in neurologic injury whether it results from ischemia, trauma both cumulative or as a single event, or a gradual degenerative process such as Alzheimer’s and Parkinson’s diseases, ALS, and multiple sclerosis. Nevertheless, even within these few mechanisms there is potential for delineation of strategies for neuroprotection and neuroregeneration or neurogenesis. 

It also must be understood that, given the complexity of these systems, a single intervention or drug is a woefully naïve approach and not a reasonable strategy to effect a therapy for these pathologies. For this reason, this paper proposes to implement a multifactorial therapy to this complex multivariate system. This therapy is designed to:

  1. scavenge free radicals resulting from injuries whether gradual or sudden
  2. protect healthy cells from neurotoxins released from damaged neurons or glia
  3. suppress inflammatory responses resulting from the injury or process
  4. to enhance cell to cell communication at the membrane lipid rafts
  5. to promote neurogenesis.

To accomplish this, a combination of naturally occurring supplements is proposed. This combination includes vitamins and more importantly, substances which are endogenous to the brain (occur naturally in the brain) and are essential for membrane structure, necessary for mitochondrial respiration, and are required to direct stem cell differentiation toward neurogenesis and myelogenesis.

It is imperative, therefore, first to understand the mechanisms and the sequence of the pathologic cascade in order to target specific places where an intervention could interrupt this sequence and thereby halt its progression.” 

It is with this introduction, the formula of Neupanex® is proposed by Dr. Clarke. Each of the components individually has been shown in countless studies, published in reputable biochemistry, neuroscience and clinical scientific peer-reviewed journals, to accomplish neuroprotection and/or neuroregeneration by one or more of the above mechanisms or interventions. Activation of free-radical scavenging microglia, driving neurogenesis via the Sigma-1 receptors, blocking inflammatory cytokines, blocking receptors for the injury-induced release of neurotoxins are all functions of the various components of Neupanex®.

Clinically, many of Dr. Clarke’s patients have benefited from Neupanex® over the past 20 or more years. Cognitive and socialization improvements in dementias and improvements in motor deficits and paralysis have all been demonstrated. In addition, because it is a packet of powder that can be mixed into a drink or shake, it can be administered via a gastrostomy tube to patients who cannot swallow due to their injury. It can be used in athletes as an aide to neuroprotection, especially after a concussive injury.

Dr. Clarke summarizes, “There is no single medication or substance that can fix neurologic injury. There is no single pill or cream that will be discovered to repair the brain. This is not like ‘penicillin kills strep throat.’ Like a symphony, the brain requires innumerable natural chemical reactions, all at the same time, to do clean-up and repair.
